From June 23 to 25, 2011, the Mesothelioma Applied Research Foundation will hold an annual international symposium on malignant mesothelioma. The Mesothelioma Applied Research Foundation is a non-profit organization, which provides funds research and services to patients that have mesothelioma.  The three-day conference will allow participants to express their concerns regarding mesothelioma research and  the ban of the use of asbestos.

The symposium will also educate attendees about the effects of asbestos to health and the existence of mesothelioma, as well as the threat it brings to public and economy. Experts from across the world will share their stories with people in the same situation. Participants can register for the conference via the organization’s website. The website will also contain the agenda for the conference starting in March.

Mesothelioma, is a rare and aggressive form of cancer that claims nearly 3,000 lives in the United States alone.  Mesothelioma is caused by the inhalation of asbestos, a fire retardant and insulator that was used widely until the 1970's.  However, there are no early symptoms and it can take over 30 years to diagnose mesothelioma. Most victims do not realize they have the disease until it is too late. Due to this lag in diagnosis time, workers in the construction, mining, industrial and even family members of those who work in these fields are still dying from the asbestos-caused disease. Household products, including children’s toys can also contain traces of the dangerous carcinogen.

Mesothelioma treatments may benefit from new research

New cancer research may prove helpful in the fight against mesothelioma. Mesothelioma is a rare cancer caused almost solely by toxic asbestos fibers. These fibers, once inhaled, can start a mutation process in cells of lung and abdominal cavity lining. Mesothelioma can take decades to demonstrate symptoms making it hard to detect. Once it has been diagnosed, typical prognoses include an average life expectancy of eighteen months.

Although many mesothelioma patients are choosing palliative care, which focuses on pain management and bettering quality of life, there are many aggressive mesothelioma treatments available. A combination of surgery, chemotherapy, radiation and rehabilitative therapies are typically administered in a mesothelioma case. However, mesothelioma has proved somewhat chemotherapy resistant and continues to be considered incurable.

As part of the Patient Protection and Affordable Care Act of 2010, grants have been distributed to further cancer research. Agios Pharmaceuticals, a recipient of such a grant through the Qualifying Therapeutic Discovery Project Program (QTDP), has been studying cancer metabolism. Following an exciting discovery of a connection between slowed growth in lung cancer cells and the diabetic drug metformin, cancer metabolism testing is proving positive.

With $488,000 in funding from QTDP, Agios Pharmaceuticals is one of the leading groups in metabolism research. Chief executive David Schenkein said, “Nutrient supply and deprivation is becoming potentially the next big wave.”

Cornerstone Pharmaceuticals, Inc., is another company to receive funding through QTDP for cancer treatment research. Cornerstone Pharmaceuticals said, “Just as the body needs nutrients to survive, cancer cells depend on certain nutrients to make energy and to proliferate.”

Until now, typical cancer treatment has focused on removal of the malignancy and attack on remaining cancerous cells. This approach often leaves surrounding healthy tissues vulnerable to the same attack, allowing patients’ quality of life to suffer as well

Cancer metabolism research is taking a new approach. Like other living organisms, cancer needs an energy source to continue growing. Glucose is a primary food, or energy source, for cancer. Researchers are testing the potential of cutting of a cancer’s glucose supply in hopes this will render the cancer unable to continue growing and spreading.

New cancer research is welcome as mesothelioma cases in America and worldwide are thought to be rising. Three thousand Americans suffer with mesothelioma each year, while the numbers worldwide are believed to be as much as ten times higher. The World Health Organization estimated ninety thousand mesothelioma cases as we head into what may be the peak of this fatal disease.

Gene therapy research proves promising

Researchers in Baltimore have made a significant discovery concerning the behavior of mesothelioma in varying patients. Their findings may help them to predict patient responses to the disease, and possibly even control those responses in order to improve survival in those with malignant peritoneal mesothelioma.

Peritoneal mesothelioma refers to the formation of cancerous tumors in the mesothelial tissues of the abdominal cavity. Like other forms of mesothelioma, the disease is caused almost exclusively by asbestos fibers and is terminal in the overwhelming majority of cases. Unlike better known forms of the disease such as pleural mesothelioma, or mesothelioma of the lung lining, the mechanism which introduces the asbestos fibers to the area of the tumor is unknown. Some evidence suggests that peritoneal mesothelioma may result from the accidental ingestion of asbestos fibers, whereas pleural mesothelioma results from inhalation of the dangerous materials.

Patients suffering from peritoneal mesothelioma react to the disease in very different ways, a fact which lead researchers to further investigation. According to the Maryland researchers, “There is marked variability in its clinical behavior. Some patients die rapidly, and others survive for many years.”

The researchers believe that the differences in individual responses to the disease may lie in the expression of certain genes which affect the chemical “signals” transmitted to cells. In some patients, these signaling pathways cause cancerous tumors to grow and spread more rapidly, while in other patients the tumors appear to grow far slower. The researchers analyzed forty-one tissue samples from mesothelioma patients, and found that the “signaling chemicals” responsible for RNA production and protein synthesis differed between samples.

In one group the over-expression of certain pathways known as PI3K and the mammalian target of mTOR caused the presence of specific genes associated with a peritoneal mesothelioma survival of around 24 months. In patients where the same pathways and the genes which they are linked to were not present, survival rates were more than twice that time at some 69.5 months. When synthetic means were introduced to inhibit the expression of the PI3K and mTOR pathways, they found that the division and growth of cancerous cells was significantly impeded.

The scientists believed that further research into the significance of specific gene expression in peritoneal malignant mesothelioma patients was warranted. They asserted that the experiments already conducted highlight the importance of analyzing gene expression and further understanding the mechanisms that link these expressions with differing survival rates.

The study concluded: “Targeting the PI3K and mTOR signaling pathways may have significant therapeutic value in patients with MPM.”
